Themonoclonalantibody1C7reactshighlyspecificwithhumannaturalandrecombinantLBP.CrossreactionswithLBPofotherspeciesarenotobserved.TheantibodybindspoorlytoLBP-LPScomplexes:itinteractswiththeLPSbindingsite.LPSbindingprotein(LBP)isanapproximately60kDaacutephaseproteinthatisproducedbyhepatocytes.ThisproteinstronglybindstoLPSandhasbeenshowntoplayanimportantroleinthehandlingofLPSbythehost.AnumberoffunctionsofLBPhavebeenreported.First,LBPtransfersLPStotheLPSreceptorCD14onmononuclearphagocytes,leADIngtoan100-1,000foldincreasedsensitivityofthecellstoLPS.FurThermore,LBPcanenhancetheresponseofCD14negativecellsbyaccelerationofLPSbindingtosolubleCD14,acomplexthatstimulatesthesecells.Next,LBPtransfersLPSintoHighDensityLipoprotein(HDL),whicheffectivelyneutralizesitsBIOLOGicalpotency.LBPwasdemonstratedtoprotectmicefromsepticshockcausedbyLPSorgramnegativebacteria.
