Description:SLPI,Human,mAb31,biotinylated
Themonoclonalantibody31recognizessecretoryleukocyteproteinaseinhibitor(SLPI).SLPIwasidentifiedasanalarmreactantandexpressionisinducedbyinflammatoryfactorslikeLPS,IL1β,TNFαandneutrophilselastase.SLPIisarelativelysmallbasicantiproteaseof11.7kDaandisacationicnon-glycosylatedproteinconsistingof107aminoacids.SLPIhasahighaffinityfortheneutrophilserineproteinases,elastaseandcathepsinG.OrthologuesofSLPIhavebeenfoundinmice,rate,pigsandsheep.ItconsistsoftwohighlysimilarWAP(‘wheyacidprotein’)/four-disulphidecoredomains.SLPIcontain16cysteineresidueswhichassembleintoeightdisulphidebridges(fourineachWAPdomain).SLPIisconstitutivelyexpressedatmanymucosalsurfacesandisproducedbyavarietyofepithelialcells,includingrespiratory,intestinalandamnioticepithelia.Expressionisalsodetectedinmastcells,neutrophilsandmacrophages.ExpressionofSLPIgeneissignificantlyincreasedbyprogesteroneandbythepro-inflammatorycytokinesTNF-αandIL1-β.AlthoughSLPIhasbeenshowntoinhibitaspectrumofproteases(includingHNE,cathepsinG,trypsin,chymotrypsinandchymase),itsmainactioninthisregardislikelytobetheinhibitionofelastase,asindicatedbyitslowdissociationconstantandfavourablekineticsofinhibitionforthisenzyme.SLPIhasbeendescribedinseveralbodyfluidslikeseminalfluid,bronchialfluids,cervicalfluidsandsaliva.Ithasbeenfoundtobeantibacterial,antifungal,anti-retroviral,andtohaveanimportantroleinmucosaldefence.SLPImightalsofacilitatetumorspread,contributingtowoundhealing,iselevatedinsepsisandlevelsseemtocorrelatewithoralcandidiasisinHIV-1positivepatients.Thereactivityoftheantibody31withisolateddomainsofSLPIwasevaluatedusingdomainsobtainedbycleavageusingpartialacidichydrolysis.Therefore,monoclonalantibody31recognizesalsootherSLPIcleavageproducts.
