ThemonoclonalantibodyBV4recognizeshumanbeta3integrinsubunitpresentinPlateletglycoproteinGPIIb-IIIa(integrinalphaIIb/beta3,CD41/CD61)andinthevitronectinreceptor(integrinalphaV/beta3,CD51/CD61).Interginsareafamilyofheterodimericmembraneglycoproteinsexpressedondiversecelltypeswhichfunctionasthemajorreceptorsforextracellularmatrixandascell-celladhesionmolecules.AsadhesionmoleculestheyplayanimportantroleinnumerousBIOLOGicalprocessessuchasplateletaggregation,inflammation,immunefunction,woundhealing,tumourmetastasisandtissuemigrationduringembryogenesis.Inadditionintegrinsareinvolvedinsignalingpathways,transmittingsignalsbothintoanoutfromcells.Allintegrinsconsistoftwonon-covalentlyassociatedsubunits,alphaandbeta.Atleast12differentalphasubunitsand8betasubunitshavebeenidentified.Thebetasubunitsallcontain56conservedcysteines(exceptbeta4whichhas48)whicharearrangedinfourrepeatingunits.Thebeta3subunitisa93kDaproteinthatcontainsalargeloopintheN-terminusstABIlizedbyintrachaindisulphidebondingwiththefirstcysteine-richrepeat.
PlateletglycoproteinGPIIb-IIIaisexpressedonplateletsandmegakaryoblasts.Itisconstitutivelyexpressedandbecomesactivatedontriggeredplatelets.PlateletglycoproteinGPIIb-IIIabindstofibrinogen,fibronectin,vWF,vitronectinandthrombospondin.Nexttothisitisalsoareceptorforseveralsolubleadhesiveproteins.Vitronectinreceptorisexpressedonendothelialcells,someBcells,monocytes/macrophages,plateletsandtumourcells.Vitronectinreceptorbindsnexttovitronectintofibrinogen,vWF,thrombospondin,fibronectin,osteopontinandcollagen.Defectsinhumanbeta3integrinareacauseofGlanzmannthrombasthenia,whichisanautosomalrecessivedisordercharacterizedbymucocutaneousbleedingandtheinabilityofthisintegrintorecognizemacromolecularorsyntheticpeptideligands.
