Description:VE-Cadherin,Human,mAbBV9,FITC
ThemonoclonalantibodyBV9bindstotheextracellulardomain(EC3-EC4)ofhumanVE-cadherin(vascularendothelialcadherin).Endothelialcellscontrolthepassageofplasmaconstituentsandcirculatingcellsfrombloodtotheunderlyingtissues.VE-cadherinisofvitalimportanceforthemaintenanceandcontrolofendothelialcellcontacts.MechanismsthatregulateVE-cadherinmediatedadhesionareimportantforthecontrolofvascularpermeABIlityandleukocyteextravasation.VE-cadherinregulatesvariouscellularprocessessuchascellproliferationandapoptosisandmodulatesvascularendothelialgrowthfactorreceptorfunctions.Therefore,VE-cadherinisalsoessentialduringembryonicangiogenesis.ThespecializedfunctionofVE-cadherinislostorimpairedinseveralpathologicalconditions-includinginflammation,sepsis,ischemiaanddiabetes-whichleadstosevere,andsometimesfatal,organdysfunction.FurThermore,abnormalincreaseinvascularpermeabilityisoftenobservedinpathologicalconditions,suchastumor-inducedangiogenesis,maculardegeneration,allergy,andbrainstroke.
Endothelialpermeabilityisregulatedinpartbythedynamicopeningandclosureofcell-celladherentjunctions.Invascularendothelium,adherentjunctionsaremainlycomposedofVE-cadherin,anadhesivereceptorthatisabletoself-associateatendothelialcellcellcontacts.VE-cadherinlinksendothelialcellstogetherbyhomophilicinteractionsmediatedbyitsextracellularpartandassociatesintracellularlywiththeactinCytoskeletonviacatenins.VE-cadherinbelongstothecadherinsuper-familyofcellcelladhesionmolecules,whichareencodedbymorethan200genesinthehumangenome.ClassicalcadherinsareCa2+-dependent,homophilic,celltocelladhesionmoleculesexpressedinnearlyallcellswithinsolidtissues.Cadherinsformacoreadhesioncomplexthatconsistsofacadherindimer,bindingthroughitsextracellularregiontoanotherdimerofcadherinsexpressedinadjacentcells,whileitsintracellularregionisanchoredtotheplasmamembraneandlinkedtothecytoskeleton.TheVE-cadherinextracellulardomainconsistsoffivecadherin-typerepeats,calledEC(extracellularcadherin)domainsthatareboundtogetherbycalciumionsinarod-likestructure.
