Themonoclonalantibody60.11recognizesacellmembraneC1qbindingmoleculethatrecognisestheglobularheadsofC1q.Itisalsopresentinplasmaandtheextracellularmatrix.Themoleculeisanunusuallyacidic,singlechainproteinwithanapparentmolecularweightof33kDa.ItdoesnotpossessaconventionalsequencemotifcompatIBLewithamembranespanningsegmentnoraconsensussiteforaGPIanchor.gC1q-Rmigratesasasinglechainunderbothreducingandnon-reducingconditions,butitbehavesasanoligomerongel-filtrationinnon-dissociatingconditions.ItsmultimerformationmaybeacriticalprocessbywhichthegC1q-RmoleculeincreasesitsaffinityformultivalentligandssuchasC1q.
gC1q-RhasbeenshowntoinhibitcomplementactivationbypreventingthebindingofC1qtoantibodies,suggestingthatthebindingsiteforgC1q-Randthebindingsiteforimmunecomplexes,whicharepresentontheC1qglobular‘heads’,maybelocatedatthesameposition.gC1q-Riscapableofinteractingwithseveralproteinsinvolvedinbloodclotting,namely,thrombin,prothrombin,theheparinbindingformofvitronectin,theternarycomplex,vitronectin-thrombin-antithrombin,aswellashigh-molecular-weightkininogenandHagemanfactor.Besidesitsroleinthecomplementpathway,gC1q-RparticipatesinenhancementofFc-receptorandCR1-mediatedphagocytosis,procoagulantactivityonplatelets,andchemotacticactivityonmastcells,eosinophils,neutrophils,andfibroblasts.gC1q-Risexpressedonawidevarietyofcellsandcanserveasabindingsiteforplasmaandmicrobialproteins.ItsantigenicsitesmaybecrypticoncellsinsUSPensionbutbecomeexposedwhenthecellsarefixedbybifunctionalcross-linkers.Probablyitisalsoexpressedonthecellmembraneasatetramer.CrosslinkingoractivationmaythusbringaboutatetramericassemblyofgC1q-Rfollowedbyaconformationalchangewithinthemolecule,therebyexposingepitopeswhichareotherwisehidden.AformofGC1q-Risalsofoundinsidethecell.IntracellulargC1q-Rhasbeenshowntobindthecytoplasmictailoftheα1B-adrenergicreceptorandtoPKCμ.
Themonoclonalantibody60.11isdirectedagainstepitopessituatedwithintheNH2-terminalstretchofgC1q-Rcorrespondingtoresidues76-93.Clone60.11recognizestheputativeC1qbindingsiteandreactswiththematureform,buthaspoorornoreactivitywiththetruncatedform,lackingresidues74-95.
