Themonoclonalantibody52B83reactswithtumornecrosisfactoralpha(TNF-alpha).TNF-alphaisahomotrimeric17kDaprotein,thatinteractswitheitheroneofthetwotypesofTNF-receptors,termedIandII,leADIngtoreceptorcross-linkingandsignaltransduction.Thereceptorsdifferstronglyintheirintra-cellularsignalingpathways.
TNF-alphawasoriginallydescribedasahighlycytotoxiccytokinefortumorcells,itcausestumornecrosisinvivoandshowscytolyticactivityagainsttumorcellsinvitro.FurThermore,-TNF-alphaisfoundtobeacentralmediatorinmanyinflammatoryandimmunologicalprocesses.Itcanbeinducedbyvariousproductsofmicro-organismsandbyvariouscytokinesleadingtoexpressionofawidevarietyof-cytokines.Thepro-inflammatorypropertiesof-TNF-alphaplayacentralroleinseveralauto-immunediseasessuchasrheumatoidarthritisandinhibitionbyneutralizingmoleculeshavebeenshowntobebeneficialinpatients.
TNF-alphawasoriginallydescribedasahighlycytotoxiccytokinefortumorcells,itcausestumornecrosisinvivoandshowscytolyticactivityagainsttumorcellsinvitro.FurThermore,-TNF-alphaisfoundtobeacentralmediatorinmanyinflammatoryandimmunologicalprocesses.Itcanbeinducedbyvariousproductsofmicro-organismsandbyvariouscytokinesleadingtoexpressionofawidevarietyof-cytokines.Thepro-inflammatorypropertiesof-TNF-alphaplayacentralroleinseveralauto-immunediseasessuchasrheumatoidarthritisandinhibitionbyneutralizingmoleculeshavebeenshowntobebeneficialinpatients.
