PolyclonalantibodyHP9058recognizeshumancomplementfactorHrelatedprotein4(FHR4).Thecomplementsystemisthefirstlineofdefenseagainstpathogensandfacilitateseliminationofapoptoticanddamagedcells.ComplementfactorH(fH)isanabundantplasmaproteinthatnegativelyregulatesthealternativepathwayandtheC3bamplificationloop.TheproteinsencodedbythefHgenefamilyincludesbesidesfHalsofivefH-relatedproteins(FHR1-5),encodedbycloselylinkedbutuniquegenes.LikefHtheseproteinsconsistofrepeatingshortconsensusrepeats(SCRs).fHcompeteswithfBforbindingtoC3bandC3b(H2O)therebyblockingtheformationoftheconvertase,acceleratesdecayofC3-convertasesandisacofactorforfI.ThefunctionofFHRproteinsislessclear.FHRsareproducedprimarilyintheliverandsecretedintheplasma.FHR4bindstotheC3dregionofC3b.IncontrasttofH,theFHR4-C3binteractionpromotescomplementactivation.TwotranscriptsoftheFHR4geneexist:FHR4BandFHR4B,consistingofrespectively9and5SCRs(86&42kDa).BothvariantshavedifferentbindingprofilestoC3b.SuggestedligandincomplementactivationofFHR4ispentamericCRP.
