ThepolyclonalantibodyrecognizesratC1q,a459kDamoleculeconsistingofthreeindividualpolypeptidechains.C1qcomposestogetherwithC1randC1stheC1macromolecule,thefirstcomponentoftheclassicalcomplementpathway.InteractionofimmunecomplexeswithC1qinducesaconformationalchangewithintheC1complex,whichresultsinactivationoftheclassicalpathway.C1qfunctionsasrecognitionunitbybindingtotheheavychainofIgGorIgM(FcgammaandFcmicro)providedthattheimmunoglobulinsareboundtotheirantigen.FurThermoreC1qcanbindtoapoptoticblebs,whereitactivatestheclassicalcomplementpathwayandmediatesphagocytosis.Assuch,C1qpromotestheclearanceofapoptoticcellsandsubsequentexposureofautoantigens,therebypreventingstimulationoftheimmunesystem.C1qispredominantlyproducedbymacrophagesbutalsobyfolliculardendriticcells,interdigitatingcellsandcellsofthemonocyte-macrophagelineage.C1qdeficiencyhasaprofoundeffectonhostdefenceandclearanceofimmunecomplexes.AbsenceofC1qmaycauseautoimmunitybyimpairmentoftheclearanceofapoptoticcells.InheritedC1qdeficiencyisalsoassociatedwiththedevelopmentofsystemiclupuserythematosus(SLE).
