Thepolyclonalantibodyrecognizes3-chlorotyrosinespecificproteinadductsthatareformedbyhypochlorousacid.PolymorphonuclearleukocytesorneutrophilsareoneoftheprimarydefensemechanismsagainstinvADIngmicroorganismsandalsoinvolvedintheremovalofnecroticcells.However,incertainconditions,recruitmentofneutrophilsmayaggravateexistinginjury.Forexample,neutrophilsareinvolvedinthepathophysiologyofhepaticischemia-reperfusioninjury,endotoxin-andsepsis-inducedliverfailure,alcoholichepatitis,andcertaindrugtoxicities.Thefactthatneutrophilsaccumulateintheliverdoesnotnecessarilymeanthattheycauseinjury.ICAM-1andCD18integrinsareinvolvedintheadherenceofneutrophilstotargetcells,e.g.,hepatocytes.Thisresultsinalong-lastingadherence-dependentoxidantstress,whichisamajorfactorinneutrophil-mediatedlivercellkilling.Stimulatedneutrophilsreleaseoxidants,proteases,andotherpotentiallyinjuriousconstituents.TheygeneratesuperoxideradicalsandhydrogenperoxideandreleaseMPO,whichcatalyzesoxidationofchloridebyhydrogenperoxidetogivehypochlorousacid.Thishypochlorousacidcanreactwithproteinstoform3-chlorotyrosineproteinadducts.AlimitationforinvestigatingthepathophysiologicalroleofneutrophilsinvivoisthelackofareliablebioMarkerforneutrophilcytotoxicity.Immunohistochemicalstainingofliversampleswithanti3-chlorotyrosineantibodycorrelateswellwithneutrophil-inducedliverinjury.Thepolyclonalantibodycanbeusedtoassesschlorotyrosineproteinadductformationandisausefulmarkerofneutrophil-inducedlivercellinjuryinvivo.
