ThepolyclonalantibodyrecognizesthehumansurfactantproteinB(SP-B).Therearefoursurfactant-specificproteins,designatedsurfactantproteinA(SP-A),SP-B,SP-CandSP-Drespectively.SP-AandSP-Darehydrophilicsurfactantproteinsandaremembersofthecollectinfamily.SP-BandSP-Carehydrophobicsurfactantproteinsandmaybethemostappropriateindicatorsfortheevolutionaryoriginofsurfactant.SP-BissynthesizedbythealveolartypeIIepithelialcellsasa40-42kDprecursorthatissubsequentlyproteolyticallyprocessedto7.8-8kD.SP-BenhancesthespreADIngandstABIlityofsurfactantphospholipidsinthealveolus.SP-Bisessentialforair-breathinginmammalsandisthereforelargelyconserved.SP-Bcaninteractwithbothphospholipidheadgroupsandfattychainsandisparticularlyactiveinenhancingsurfaceactivebehaviourinendogenousandexogenouslungsurfactants.EvenlowSP-Bcontentshadmeasurableeffectsinincreasingtheadsorption,dynamicsurfacetensionlowering,and/orfilmrespreadingofDPPC,mixedsyntheticlipids,andcolumn-purifiedlungsurfactantphospholipids.DeficiencyofSP-Bandothersurfactantcomponentsisassociatedwithrespiratorydistresssyndrome(RDS)inprematureinfantsandadultswithrespiratorydistresssyndrome(ARDS).ThepolyclonalantibodyisraisedagainstpigSP-Bfrompiglungswhichhasbeenreconstitutedinmicellesoflysophosphatidylcholine.TheantibodyrescognizesundernonreducingconditionsthemostprominentformsofSP-Bof8kDand22kD,correspondingtoSP-Bdimers.ThepolyclonalantibodyiscrossreactivewithhumanSP-B.
