ThepolyclonalantibodyrecognizestheextracellularpartofthemouseTumorNecrosisFactorReceptortype2(TNF-RII)ofthemembrane-boundaswellasthesolublereceptor.TNF-RII(~75-80kDa)ispresentonmostcelltypesandisconsideredtoplayaprominentroleincellstimulationbyTNF-alpha.TNF-alphaactivatesinflammatoryresponses,inducesapoptosis,regulatescellularproliferation,andmayevenpromotecancerprogression.TheeffectsofTNF-alphaaremediatedbyTNF-RIandTNF-RII,whichhavebothdistinctandoverlappingdownstreamsignalingcascades.InductionofcytotoxicityandotherfunctionsaremediatedlargelyviaTNF-RI.TNF-RIisequallywellactivatedbyboththe17kDasolubleand26kDamembrane-boundform,whereasTNF-RIIisefficientlyactivatedonlybythemembraneboundformofTNF-alpha.BindingoftheinherentlytrimericTNF-alphatoTNFR1andTNFR2inducesreceptortrimerizationandrecruitmentofseveralsignalingproteinstothecytoplasmicdomainsofthereceptors.OccupancyofTNFR2resultsindirectrecruitmentofTNFReceptorAssociatedFactor2(TRAF2),whichinturnrecruitsTRAF1.
