AntibodiesderivedfromcloneMaP.mGILT6recognizemousegamma-interferon-inducIBLelysosomalthiolreductase(GILT).GILTfacilitatesmajorhistocompatibilitycomplex(MHC)classII-restrictedprocessingthroughreductionofproteindisulfidebondsintheendocyticpathway.GILTcanalsofacilitatethetransferofdisulfide-containingantigensintothecytosol,enhancingtheircross-presentationbyMHCclassI.HumanGILTconsistsof261aminoacidswitha37aminoacidsignalsequenceanda224aminoacidprecursorform.Ingeneral,the35kDaprecursoristaggedwithmannose-6-phosphate(M6P)residuesandtransportedtotheendocyticpathwayviatheM6Preceptor(M6PR).Asmallfractionoftheprecursorissecretedasadisulfide-linkeddimer.Inearlyendosomes,N-andC-terminalpro-peptidesarecleavedtogeneratea28kDamatureform.ThematureformofGILTislocalizedtolateendosomesandlysosomesandhasmaximalreductaseactivityattheacidicpHfoundinthesecompartments.GILTisconstitutivelyexpressedinprofessionalantigenpresentingcells(APC).TheconstitutiveexpressionofGILTinAPCsislikelytoaccountforenhancedreductionofproteinsinthelateendosomal,lysosomal,andphagosomalcompartments.AcombinationofGILT-mediatedreductionandproteolysisinthethesecompartmentscanenhancethegenerationofclassIIepitopesaswellasfacilitatethetranslocationofinternalizedantigensintothecytosolforproteosomalprocessingandcross-presentation.Itisalsoconstitutivelyexpressedinthymocytes,matureTcells,andsomefibroblasts.IFN-ɣplaysisthekeyfactorininducingexpressionofMHCclassIIandothercomponentsoftheclassII-restrictedprocessingpathway.IFN-ɣsignalsthroughaspecificcellsurfacereceptorfollowedbyactivationofJAK/STAT1signalling.
