Monoclonalantibody-cloneM7/1recognizeshumanendothelial-monocyte-activatingpolypeptideII(EMAPII).EMAPIIisaproinflammatorycytokineandchemoattractantofmacrophagesandpolymorphonuclearlymphocytes.TheproteinwasinitiallyidentifiedasaproductofmurinemethylcholanthreneA-inducedfibrosarcomacells.EMAPIIissynthesizedasa34kDaprecursormolecule(proEMAP),-itcanrunanomalouslyinSDSpageat43kDa,andisenzymaticallycleavedtoproduceaBIOLOGicallyactive22kDamaturepolypeptide.Itmodulatesarangeofpropertiesofendothelialcells,monocytes,andneutrophilsinvitro,andinducesanacuteinflammatoryreactionandtumorregressioninvivo.Lateritwasfoundtobeanti-angiogenicandcapableofinductionofapoptosisinproliferationandhypoxicendothelialcellsinvitrointumorvasculatureinvivo.EMAPIIisreleasedfromcells,byayetunknownmechanism,aspro-andmatureEMAPIIproteinsinresponsetovariousformsofcellularstresses,includingglucosestarvationandhypoxia.Endothelialcellsarethemaintarget-forEMAPIIandexplaintheanti-angiogenicproperty.Theanti-tumoreffectisexpectedtoalargeextenttobemediatedthroughthischaracteristic.EMAPIIisalsoassociatedwithbraindiseasesandinjury.EMAPIIexpressionisrelatedwithleukocytes-infiltratedinflamedtissueandsubsequentapoptosis.Insolidtumorsthereisalsoanimmunosuppressiveroleidentified.EMAPIIinducingtumorcellsinduceapoptosisoflymphocytes.EMPAIIisconsideredaMarkerformicroglialcellsandmacrophagesinbraindiseases.ThedistributionofEMAPIIanditsprecursorinbiopsiesmightbeapplicableasadditivediagnostictool.
