Themonoclonalantibody5B8reactswithmouseosteoclast-associatedreceptor(OSCAR),whichisa265aminoacidtype1transmembraneproteinbelongingtotheleukocytereceptorcomplexproteinfamilythatplaycriticalrolesintheregulationofbothinnateandadaptiveimmuneresponses.Inmice,OSCARisaRANKL-inducIBLegenewhich-isexpressedduringthelaterstagesofpre-osteoclastmaturationandisshowntobeapotentFcRγ-associatedcostimulatoryreceptorinvitro.OSCARpromotesosteoclastogenesisinvivo,andOSCARbindingtoitscollagenligandledtosignalingthat-isinvolvedinfunctionaldifferentiationofleukocytes.InrecentstudieshumanOSCARhasbeenshowntoplaymultiplerolesintheimmunesystemincludingactivationandmaturationofimmunecells,preventionofapoptosisandenhancementofpro-inflammatorycircuits.Thelatterby-inducingintracellularcalciumreleaseresultinginsustainedIL-8andchemokineexpressionleADIngtochemotaxisofTh4effectorandregulatoryT-cells.Ina-murinemodelusingcalvariasboneexplants,thesynovialfluidofosteoarthritispatientsresultedinincreasedOSCARmRNAexpressionsuggestingthatOSCARmightcontributetointhedegenerativeprocess.Moreover,inarecentstudy-OSCARwasfoundtobe-inducedinmonocytesofRApatients,facilitatingtheirdifferentiationintoosteoclastsandboneresorption.Themonoclonalantibody,5B8,canbeusedforanalysisofmouseOSCARinflow-cytometryaswellasfunctionalstudiesforosteoclastogenesis.
