Monoclonalantibody1A2recognizesmousecomplementfactorH(CFH).CFHisthefirstregulatoryproteinofthealternativepathwayofthecomplementsystem.Therearethreepathwaysofcomplementactivation;classical,alternative,andlectinactivationpathway.ThesepathwaysconvergetoformC3/C5convertasesthatgenerateC3a,C3b,C5a,andC5b,eachwithsubstantialBIOLOGicactivity.Complementregulatorsarenecessarytopreventtheinjudiciousproductionofthesemediatorsandpotentialinjurytoself-tissue.TheplasmaproteinsCFH,C4-bindingproteinandthecellmembraneproteinscomplementreceptor1(CR1;CD35),decay-acceleratingfactor(CD55),andmembraneco-factorprotein(CD46)allaremembersoftheregulatorsofcomplementactivationfamily.TheseproteinshavenaturalaffinityforC3band/orC4b,whichconfersonthemtheABIlitytoacceleratetheintrinsicdecayofC3/C5convertasesand/oractasco-factorforthecleavageandinactivation(i)ofC3bandC4bbycomplementfactorI(CFI).GenetichumananalysesrevealsaclearassociationofCFHwithdifferenthumandiseases.Theseincludediseasesofthekidney,theatypicalformofHemolyticUremicSyndrome(aHUS)andmembranoproliferativeglomerulonephritis(MPGN).FurThermore,CFHisassociatedwithage-relatedmaculardegeneration(AMD),adiseaseoftheeye.
