Themonoclonalantibody7A6(previouslyknownasmCD59.3)recognizesmouseCD59a,apotentinhibitorofthecomplementmembraneattackcomplex(MAC)action.CD59regulatestheformationandfunctionofthelyticC5b-9complexbybindingC8andpreventingtheunfoldingandmembraneinsertionofC9andbybindingC9andrestrictingitspolymerization.CD59isasmall(18-25kDa)molecule,linkedtothecellmembranethroughaglycosylphosphatidylinositol(GPI)anchorandcomprising77aminoacidswithasingleN-linkedcarbohydrategroupatAsn-18.AnaloguesofCD59canbefoundinallspecieswithsimilarstructuresandsizes.
Incontrasttoallotherspecies,micehavetwogenesencodingaCD59aandaCD59bprotein.Thesetwomoleculesare63%identicalattheaminoacidlevelandshareallmajorstructuralfeatures.CD59aisbroadlydistributedonendothelia,erythrocytes,plateletsandonnumerousothercelltypesinorgans,adistributionpatternresemblingthatofCD59inotherspecies.ExpressionofCD59bisrestrictedtogermcellelementsinthetestisandmaturespermatozoa.BothCD59aandCD59binhibithumanandrodentcomplementwithsimilarefficiency.CD59maybeinvolvedinrheumatoidarthritis,motornerveinjuryintheGuillain-Barrésydromeandinotherdiseaseswheredefectiveinhibitionofcomplementactivationonselftissueisinvolved.FurThermore,CD59mayplayanimportantpartinabrogatingtheeffectsofcomplementattackinrenaldisease.Itspresenceandprotectiveeffecthavealreadybeendemonstratedonhumanrenalcells.
