Themonoclonalantibody3D5recognizescomplementdecayacceleratingfactor(DAF),alsodesignatedasCD55.Cellsexpressontheirsurfaceseveralproteinswhichprotectagainstcomplementattack,namelyCreceptorI(CR1),decayacceleratingfactor,membranecofactorprotein(MCP)andCD59.CR1,CD55andMCPregulatetheactivationpathwaysofcomplementbyeitheracceleratingdecayoftheC3andC5convertase(CR1,CD55),oractingascofactorsfortheserineproteasefactorI,whichcleavesandirreversIBLyinactivatesC3b(CR1,MCP).
MouseCD55isa60kDatransmembraneproteinthatbindsC3bandC4btoinhibitformationandhalf-lifeoftheC3convertases.CD55isbroadlydistributedamongcellsincontactwithserum,includingbothhaematopoieticandnonhaematopoieticcells.AlthoughCD55doesnothaveanessentialroleincontrollinghemolysisoferythrocytes,ithasanimportantroleinregulationofthedepositionofC3onnucleatedcells.
TogetherwithothercomplementregulatorsCD55protectsself-cellsfromautologouscomplement-mediatedinjury.CD55cooperateswithCD46incircumventingautologousC3deposition,whileCD59inhibitsthepathwayatthecriticalend-point.
