Themonoclonalantibody6C7.1recognizesmousevascularcelladhesionmolecule(VCAM-1)(~81kDa),amemberofasubclassoftheimmunoglobulinsuperfamily(IgSF).IgSFmembersareligandsforintegrins.Celladhesionmolecules(CAMs)haveimportantrolesintheimmuneresponse,immunesurveillanceandcell-cellrecognition,especiallyinleukocyte-endothelialcelladhesion.CAMsonthesurfaceofleukocytesandendothelialcellsareactivelyinvolvedintherecruitmentofspecificleukocytesubsetsintodifferenttissues.VCAM-1isexpressedoninflamedvascularendothelium,aswellasonmacrophage-likeanddendriticcelltypesinbothnormalandinflamedtissue.Celladhesionmolecules,likeVCAM-1,areupregulatedoncerebralvesselsduringinflammatoryconditionsofthecentralnervoussystemsuchasexperimentalautoimmuneencephalomyelitis(EAE),amodelsystemformultiplesclerosis.AdmiNISTrationofmonoclonalantibody6C7.1hasbeenshowntoinhibitordiminishclinicalorpathologicalsignsofEAE.VCAM-1isareceptorforencephalomyocarditisvirusonmurinevascularendothelialcells.ExpressionofVCAM-1onvascularendothelialcellsisinducedbyTNF-alpha,IL-1,IFN-gammaorendotoxin.VCAM-1isaligandfortheintegrinsalpha4beta1(VLA-4)andalpha4beta7(LPAM-1).Theseintegrinsareconstitutivelyexpressedbythymocytes,lymphocytesandmonocytes.VCAM-1/VLA-4interactionmayplayapathophysiologicalroleinimmuneresponsesandaswellasinleukocyteemigrationtositesofinflammation.
