Themonoclonalantibody3F1recognizesmousepoliovirusreceptor,alsonamedCD155.CD155isexpressedoncellsofhematopoieticoriginandbyendothelialvesselsinsidesecondarylymphoidorgansincludingHEV.Uponactivation,dendriticcells,TandBcellsupregulatethesurfacelevelofCD155.CD155isan~45kDamemberofasubfamilyofimmunoglobulin-likeadhesionreceptors.Nectin1-4areothermembersofthissubfamily,functioningincell-cellcontactbyformingheterophilicandhemophilictrans-dimers.Incontrasttonectins,CD155lacksselfadhesionpotentialbutcaninteractwithothernectins.Thisway,CD155contributesintheformationofadherensjunctionsbetweenendothelialandepithelialcells.Duetothisadhesivecapacity,CD155influencesprocessessuchascellmotility,migration,growth,andformationofadherensjunctions.FurThermore,CD155isinvolvedinimmunologicalprocessessuchasCD8Tcell-mediatedtumorrejection,andNKcell-mediatedkilling.Manytumortypesdisplayanaberrantup-regulationofCD155makingthemvulnerabletodestructionbyNKorcytotoxicTcells.
Inhuman,poliovirus,vitronectinandseveralothercellularreceptorswereidentifiedasligandsthatbindtoCD155.CD96(TACTILE)andCD226(DNAM-1),bothmembersoftheIg-superfamily,areotherligandsofhumanCD155.MurineCD155doesnotserveasafunctionalpoliovirusreceptor,however,murineCD155recognizesthesamepanelofligandslikenectin-3,CD96,CD226andvitronectin.CD155-deficientmicedevelopnormally,however,areducedhumoralimmuneresponsetoantigensenteringtheorganismviathegastrointestinaltractisshown.
