Themonoclonalantibody6A6recognizesmouseCD96.CD96orTcell-ActivatedIncreasedLateExpression(TACTILE)isan~67kDimmunoglobulin-likeproteinexpressedonnaturalkiller(NK)andTcells.CD96belongstotheIg-superfamily,consistingofthreeIg-likedomainsandanextendedserine/threonine/praline-richstalkintheectodomain.CD96isupregulateduponTcellactivationandisinvolvedinengagementofdiseasedcellsandcellmovementtowardsareasofinflammation.MouseCD96bindstoCD155andnectin-1.CD96/CD155-interactionispreservedbetweenmanandmousealthoughbothreceptorsareonlymoderatelyconservedinaminoacidsequence.Themonoclonalantibody6A6completelyinhibitsbindingofCD96tobothrecombinantmouseCD155andmousenectin-1.Also,TcelladhesiontoinsectcellsexpressingmCD155isblockedbythismonoclonalantibody,however,dependingontheTcellsubtype.Themonoclonalantibody6A6wasraisedafterimmunizationofratswithpurifiedrecombinantmCD96-hIgG1.
