TheOX-2,orCD200receptor(CD200R),belongstotheimmunoglobulinsuperfamily.CD200RcontainstwoIg-likeextracellulardomainsandmediatesinhibitorysignalsinmyeloidcells.MicedeficientfortheligandCD200developenhancedexperimentalallergicencephalomyelitisandcollagen-inducedarthritis.UnlikeCD200R,theCD200R-likereceptorsfunctionsasactivatingreceptors.TheycontainshortcytoplasmictailsandalysineresidueinthetransmembraneregionandarelikelytosignalviaadaptorproteinssuchasDAP12,DAP10,FcRγ,orCD3ζ.
CD200R3isexpressedpreferentiallyoncellsofthemyeloidlineage,includingmastcellsandbasophils.Bothcelltypesareinvolvedinthehostdefensesystemagainstpathogensandinthedevelopmentofallergicdisorders.Theyfunctionasindependentessentialinitiatorsofallergicreactions.Basophilsaretheleastcommonleukocytesintheperipheralbloodaccountingapproximately0.5%ofallleukocytes.InvivodepletionofbasophilsbyusingmonoclonalantibodyprovedtheessentialroleofbasophilsinthedevelopmentandmaintenanceofIgE-mediatedchronicallergicinflammation.
TheactivatingCD200receptors,likeCD200R3,playanimportantroleinIgE-independentmastcellandbasophilactivation.MonoclonalantibodyBa91activatesbasophilsexvivoandelicitssystemicanaphylaxiswhenadmiNISTeredinvivo.Cross-linkingofCD200R3onMC/9cellsbymonoclonalantibodyBa91inducesdegranulation.MonoclonalantibodyBa91isalsousefulfordepletionofbasophils.
