ThemonoclonalantibodyVEB4-7E4recognizesmouseEphrintype-Breceptor4(EphB4),an~110kDprotein.Erythropoietin-producinghumanhepatocellularcarcinoma(Eph)receptorsandephrinsaremembraneproteins.Theyareclassifiedinto2broadsubclasses,namelyAandB,accordingtostructuralhomologiesandbindingspecificities.Ephreceptorsaretyrosinekinases,whichautophosphorylateuponbindingtotheircognateephrinligands.Ephreceptorsandephrinsarefrequentlyexpressedinreciprocalpatternsthatcorrelatewithcellularboundariesduringembryonicdevelopment.TheinteractionbetweenEphB4anditsligand,Ephrin-B2,playsanimportantroleincell-cell,cell-extracellularmatrixinteractionsaswellasincellmigration,adhesionandproliferation.Duringtheearlystagesofvasculardevelopment,EphB4isspecificallyexpressedinvenousendothelium,whereasEphrin-B2isexpressedinarterialendothelium.Inmouseembryo"s,EphB4andEphrin-B2areessentialforembryonicheartdevelopmentandangiogenesis.Inadultmicrocirculation,EphB4isnotaubiquitousMarkerofarterial/venouspolarity,butisexpressedalongbothvenulesandarterioles.FurThermore,EphB4isupregulatedbyendothelialcellsalongblind-endedcapillarysproutsversusconnectingcapillaries.Assuch,EphB4isthoughttoplayaroleinthepatterningofnewvesselsduringangiogenesis.EphB4isalsoexpressedinavarietyoftumorcells,likegastrointestinal,prostate,bladder,breast,liver,lungandovariancancers,aswellasleukemia,mesothelioma,malignantbreasttumorsandmelanoma.ReductionofEphB4activityacceleratedtumOrigenesisincolonandrectum.Inheadandnecksquamouscellcarcinomaandendometrialcarcinoma,overexpressionofEphB4isinverselyrelatedtoapoorprognosis.However,inmesothelioma,up-regulationofEphB4resultedingrowthofthetumor.BesidestheessentialexpressionofEphB4,coexpressionofotherEphB4familymembersorEphB-ligandsmayaffecttumorcellviABIlityandproliferationaswell.ThemonoclonalantibodyVEB4-7E4marksvenousendothelialcells,butnotarterialendothelialcellsinB16melanomacells.
