Themonoclonalantibody55R-170recognizestheextracellularpartofmouseTumorNecrosisFactorReceptorsuperfamilymember1A(TNF-RI),alsoknownasCD120aorp55.TNF-RIbelongstothelargeTNFreceptorfamily,amongwhichTNF-RII(TNF-Rp75-80),lymphotoxin-betareceptor(LTbetaR)andtheHerpesvirusentrymediator(HVEM).LigandsforthesereceptorsbelongtotheTumorNecrosisFactor(TNF)superfamilyofcytokines,whichactivatesignalingpathwaysforcellsurvival,death,anddifferentiationthatorchestratethedevelopment,organizationandhomeostasisoflymphoid,mammary,neuronalandectodermaltissues.TNF-RIcontainsacharacteristicstructuralcassettetermeddeathdomaininitssequencethatisconservedwithinadistinctsubsetofotherTNF-Rfamilymembers,suchasCD95,DR3,DR4,andDR5.Thisdeathdomain,wascharacterizedasbeingessentialforinductionofapoptosisinvitroandhasbeenstructurallyconservedwithintheseTNF-Rsuperfamilymembers.DeletionofthedeathdomainoftheTNF-RIresultsinanon-functionalreceptor,indicatingthatthedeathdomainisrequiredforthesignaltransductionofthephysiologicalfunctionsofTNF-RIinvivo.TNF-RIisa55kDtypeItransmembraneproteinandisexpressedonavarietyofcelltypesatlowlevels.ItisconsideredtoplayaprominentroleincellstimulationbyTNF-alpha.InductionofcytotoxicityandotherfunctionsaremediatedlargelyviaTNF-RI.TNF-RIispresentassolubleforminbodyfluids(forinstanceplasmaandCSF).ThisextracellularTNF-RIisgeneratedbytwomechanisms,namelyproteolyticcleavageofTNF-RIectodomainsandreleaseoffull-lengthTNF-RIinthemembranesofexosome-likevesicles.TNF-RIandTNF-RIIbothinteractwiththehomomericformsofLTbetaorTNF.However,TNF-RIfunctionsasthehighaffinityreceptorforsolubleTNF(sTNF).TNF-RIhasbeenshowntobeinvolvedinawidevarietyofinflammatorydiseases,amongwhichneurodegenerativediseases(Parkinson"sandAlzheimer"sdisease),multiplesclerosis,asthma,atherosclerosis,rheumatology.Themonoclonalantibody55R-170alsorecognizesthesolublereceptor..
