MonoclonalantibodymT2.4reactswithmouseToll-likereceptor2(TLR2,CD282).Toll-likereceptors(TLR)arehighlyconservedthroughoutevolutionandhavebeenimplicatedintheinnatedefensetomanypathogens.InDrosophilatollisrequiredfortheanti-fungalresponse,whiletherelated18-wheelerisinvolvedinantibacterialdefenses.Inmammals,TLRidentifiedastypeItransmembranesignalingreceptorswithpatternrecognitioncapABIlities,havebeenimplicatedintheinnatehostdefensetopathogens.TLR2hasbeenidentifiedasareceptorthatiscentraltotheinnateimmuneresponsetolipoproteinsofGram-negativebacteria,severalwholeGram-positivebacteria,aswellasareceptorforpeptidoglycanandlipoteichoicacidandotherbacterialcellmembraneproducts.AfunctionalinteractionbetweenTLR2andTLR6inthecellularresponsetovariousbacterialproductshasbeendiscovered.ThecurrentlyacceptedparADIgmregardsTLR2asanessentialreceptorformanyeubacterialcellwallcomponents,includinglipoproteinsandpeptidoglycan.Bacterialspeciesasdiverseasmycobacteria,spiRochetes,mycoplasma,Staphylococcusaureus,andStreptococcuspneumoniaehaveallbeenshowntomediatecellularactivationviaTLR2.ThemonoclonalantibodymT2.4inhibitsmurineTLR2-mediatedcellactivation.ItmasksthelipopeptidebindingsiteintherecombinantextracellulardomainofTLR2(TLR2ECD)anddoesnothinderthereceptorcomplexformation.TheantibodyreactswiththenativeTLR2proteinandnotwithdenaturedTLR2protein.FurThermoretheantibodystainsoverexpressed,aswellasendogenouscellsurface-andintracellularmouseTLR2.
