ThemonoclonalantibodyER-MP42specificallyreactswithmajorhistocompatibilitycomplex(MHC)classIantigensofthemouseand,therefore,itisavaluabletoolforstudyingcytotoxicT-cellinteractionswithclassIpositiveantigenpresentingcells.TheprimaryimmunologicalfunctionofMHCmoleculesistobindandpresentantigenicpeptidesonthesurfacesofcellsforrecognitionbytheantigen-specificTcellreceptors(TCRs)oflymphocytes.MHCclassImoleculesareexpressedonthesurfacesofmostcellsandarerecognizedbyCD8-positivecytotoxicT-cells,anessentialstepforinitiatingtheeliminationofvirallyinfectedcellsbyT-cell-mediatedlysis.MHCclassImoleculesareheterodimerscomposedofanalpha(44kD)andabeta(betamicroglobulin,11kD)subunit.Thefirsttwostructuraldomainsofthealphasubunitassociatetoformthepeptide-bindingpocket.ThemonoclonalantibodyER-MP42reactswithMHCclassI,anantigenthatisexpressedbyallsomaticcellsatvaryinglevels.Lymphocytesarehighlypositive,whereasfibroblastsandneuronsshowonlyalowlevelofantigenexpression.ER-MP42recognizesmurineMHCclassImoleculesonthesurfaceofcellsofthefollowinghaplotypes:H-2Fv,H-2Dd,H-2k,q,s.Weakerreactivityisfoundinmiceofthefollowinghaplotypes:H-2p,r,w7,w22.MHCclassImoleculesofotherhaplotypesarenotrecognizedbyER-MP42.Thereisnocross-reactivitywithhumanMHCclassImolecules.
