ThemonoclonalantibodyER-HR52specificallyreactswithmajorhistocompatibilitycomplex(MHC)classIantigensofthemouseand,therefore,itisavaluabletoolforstudyingcytotoxicT-cellinteractionswithclassIpositiveantigenpresentingcells.TheprimaryimmunologicalfunctionofMHCmoleculesistobindandpresentantigenicpeptidesonthesurfacesofcellsforrecognitionbytheantigen-specificTcellreceptors(TCRs)oflymphocytes.MHCclassImoleculesareexpressedonthesurfacesofmostcellsandarerecognizedbyCD8-positivecytotoxicT-cells,anessentialstepforinitiatingtheeliminationofvirallyinfectedcellsbyT-cell-mediatedlysis.MHCclassImoleculesareheterodimerscomposedofanalpha(44kD)andabeta(betamicroglobulin,11kD)subunit.Thefirsttwostructuraldomainsofthealphasubunitassociatetoformthepeptide-bindingpocket.ThemonoclonalantibodyER-HR52reactswithMHCclassI,anantigenthatisexpressedbyallsomaticcellsatvaryinglevels.Lymphocytesarehighlypositive,whereasfibroblastsandneuronsshowonlyalowlevelofantigenexpression.ER-HR52recognizesmurineMHCclassImoleculesonthesurfaceofcellsofthefollowinghaplotypes:H-2Db,H-2Dw16,H-2d,p,q.Weakerreactivityisfoundinmiceofthefollowinghaplotypes:H-2f,r,s,w17,w23,w27.MHCclassImoleculesofotherhaplotypesarenotrecognizedbyER-MP52.Thereisnocross-reactivitywithhumanMHCclassImolecules.
