ThemonoclonalantibodyER-HR3isreactivetothemajorityofmousemonocytesandasubsetofmatureresidentmacrophages,especiallythoselocatedinhemopoieticorgans.WhereasallotherleukocytesareER-HR3negative,upto70%ofcirculatingmonocytesstainpositivewiththismonoclonalantibody.FurThermore,about30%ofbonemarrowcellsexpresstheantigen.OtherER-HR3positivemacrophagescanbefoundinthesplenicredpulp,inthemesentericlymphoidparacortex,interfollicularareasofPeyer"spatchesandbonemarrow.FewER-HR3positivecellsareobservedinthethymiccortexandtheconnectivetissuesofthegastro-intestinaltract,thedermisandtherenalmedulla.EpidermalLangerhanscellsalsoexpresstheantigen.ThemonoclonalantibodyER-HR3canbeusedtoidentifyandlocalizeaverydistinctmaturetissuemacrophagesubpopulationfoundinvariousmouseorgansanditsdistributionpatternisuniquecomparedtothatfromothermacrophage-specificantibodies.TheantibodyisespeciallysuitableforontogenicstudiesbecauseER-HR3positivemacrophagesarecloselyrelatedtohemopoieticislands,especiallyaterythropoieticsites.
