ThemonoclonalantibodyER-TR3reactswithmousemajorhistocompatibititycomplexclassII(MHCclassII),alsoknownasI-AorI-E.MHCclassIImoleculesareheterodimersofnon-covalentlyassociatedalpha(31-34kDa)andbeta(26-29kDa)chains.MajorhistocompatibilitycomplexclassIIantigenpresentationrequirestheparticipationoflysosomalproteasesintwoconvergentprocesses.First,theantigensendocytosedbytheantigen-presentingcellsmustbebrokendownintoantigenicpeptides.Second,classIImoleculesaresynthesizedwiththeirpeptide-bindingsiteblockedbyinvariantchain(Ii),andtheyacquirethecapacitytobindantigensonlyafterIihasbeendegradedinthecompartmentswherepeptidesreside.MHCclassIImoleculespresentexogenouslyderivedantigentoCD4+Tlymphocytes,whichareusuallyThelpercells.CD4interactswithnon-polymorphicresiduesofMHCClassII.ThemonoclonalantibodyER-TR3isavaluabletoolforstudyingThelpercellinteractionwithclassIIpositiveantigenpresentingcells(dendriticcells,Bcellsandmacrophages)andforstudyingthedevelopmentofThelpercellssincetheystainstromalcellsinthymus.ThelevelofantigendetectedbyER-TR3differsfromstraintostrain.ThemonoclonalantibodyER-TR3crossreactswithhumanMHCclassIIontonsil.DistributionofER-TR3amongmousestrainswithindependentandrecombinanthaplotypes.
