MonoclonalantibodyER-TR7recognizeswithanintracellularcomponentofmousefibroblasts.Fibroblastsaretheleastspecializedcellsintheconnective-tissuefamily.Theyaredispersedinconnectivetissuethroughoutthebody,wheretheysecreteanonrigidextracellularmatrix(ECM)thatisrichintypeIand/ortypeIIIcollagen.Conectivetissueconsistsofglycosaminoglycans,proteoglycansandglycoproteinsthroughwhichvariousfibresrun.Thesefibrescanbecollagenous,elasticorreticular.Reticularfibresarecomposedfromthefamilyofcollagenproteinsandgivetensilestrength.Thesefibresaremadebyreticularfibroblasts.Theactivationoffibroblastsbyinflammatorystimuliresultsintheirmigration,proliferationanddepositionofextracellularmatrixcomponents,importantfeaturesinvolvedinbothwoundhealingandfibrosis.
TheER-TR7antigenisaubiquitouscomponentofstromal(interstitial)matrixcartilageandofatleastsomebasementmembranezones.Theantigendetectedisnotabasementmembranecomponent,noranymajorcollagentypeorfibronectin.Theantigendetectedhasawidertissuedistributionthanreticulin.ER-TR7detectsanintracellularcomponentoffibroblasts.SinceER-TR7doesnotreactwithpurifiedlaminin,collagentypesI-V,fibronectin,heparinsulfateproteoglycan,entactinornidogen,itdetectsahithertouncharacterizedantigen.
ThemonoclonalantibodyER-TR7canbeusedtostudythemicro-anatomyofvariousorgans.ER-TR7outlinesthevariouscompartmentsofperipherallymphoidorgansbycharacteristiclabelingpatterns(nosuchcompartmentsarefoundincentrallymphoidorgans).FurThermoreER-TR7delineatesvarioustypesofconnectivetissuecompartmentsinnonlymphoidorgans.
TheantibodyER-TR7detectsreticularfibroblasts,whichconstitutethecellularframeworkoflymphoidandnonlymphoidorgansandtheirproducts.ER-TR7isusefultoclearlydelineatedthefollicles,periarteriolarlymphoidsheathandmarginalzone;themajorwhitepulpcompartments.Furthermoreinlymphnodes,thecapsule,sinuses,follicles,paracortexandmedullarycordsareclearlydelineated.
