Themonoclonalantibody10/92recognizesthemousereceptorC5a(C5aR,CD88).ThemouseanaphylatoxinC5aisa74-residuglycopolypeptidewhichisgeneratedbyproteolyticcleavageofthecomplementfactorC5inthecourseofcomplementactivation.AvarietyofBIOLOGicaleffectsevokedbyC5acouldbedemonstrated,renderingthismoleculeanimportantmediatorofinflammation,withgranulocytesandmacrophagesasthemaintargetcells.AllcellularresponsestoC5aarespecificallymediatedbyinteractionswiththemembraneboundC5areceptor,aseventransmembraneGTP-binding-protein-coupledreceptorthatbelongstotherhodopsinsupergenefamily.The45kDaC5areceptorisexpressedbyneutrophils,monocytes/macrophages,dendriticcells,astrocytesandmicroglia.Incontrast,C5areceptorisnotdetectableonrestingorstimulatedmurineTandBlymphocytes.Themonoclonalantibody10/92doesnotinhibitthebindingofC5atoitsreceptor.
