Themonoclonalantibody440creactswithSiglec-H,acell-surfacereceptormoleculeselectivelyexpressedonmurinenaturalinterferonproducingcells(IPC),alsocalledmouseplasmacytoiddendriticcells(pDC).SiglecHisuniqueamongSialicacid-bindingIg-likelectins(Siglecs)proteinsbecauseitassociateswiththeadaptorproteinDAP12.DAP12recognizecertainvirusesandCpG-DNAthroughTLR9,resultinginsecretionofIFN-alpha,IL-12andproinflammatorychemokines.TogetherthesecytokinesandchemokinesrecruiteandactivateNKcellsandTcellsaswellasmodulatingtheantigenpresentingfunctionofdendriticcells(DC).IPCthemselvesalsofunctionasantigenpresentingcellsthatexpandmemoryTcellsandinduceTh1differentiation.ThereforeIPCmayprovideafirstlineofhostdefenseagainstviralinfectionsbyactivatingbothinnateandadaptiveresponsesinvivo.Themonoclonalantibody440cexclusivelyrecognizesmouseSiglec-Hinalllymphoidorgansunderbothnormalandinflammatoryconditions.Siglec-HisnormalpresentintheTcellzoneoflymphnodesandspleen.IncubationofIPCwithantibody440cinvitrooradmiNISTrationofantibody440cinvivoreducessecretionofIFN-alphainresponsetoCpGDNAwithoutcausingIPCdepletion.
