MonoclonalantibodyCRFD-2712(alsoknownasJC63.1or63)reactswithmouseCD36(85-kDa),acellsurfaceclassBscavengerreceptor.Recently,CD36hasbeenidentifiedasaproteinthatisrequiredforTLR2recognitionofdi-acylatedbacteriallipopeptidesandlipoteichoicacid.CD36isaheavilyN-glycosylatedtransmembraneproteinwithtwoshortintracellulardomainsandalargeextracellulardomain.CD36expressionisprimarilycontrolledbythetranscriptionheterodimerPPARg-RXR(peroxisomeproliferator-activatedreceptor-g-retinoid-X-receptor).CD36ispreferentiallyfoundwithinlipidrafts,whichfacilitatesitsassociationwithreceptors,signallingandadaptormolecules.CD36isexpressedonmonocytes,macrophages,endothelialcells,hepatocytes,erythrocytes,ADIpocytes,cardiomyocytesandredskeletalmuscle.Importantlyitisexpressedbysteriodogeniccellsintheadrenalgland,testisandovary.CD36hasbeenimplicatedinmanyBIOLOGicalprocessesincludingangiogenesis,phagocytosis,inflammation,andlipidandglucosemetabolism.CD36isassociatedwithSrc-familykinasesandwiththeintegrinsalpha3beta1andalpha6beta1.Itisareceptorandtransporterofoxidizedlipidsandlongchainfattyacids.CD36hasbeenshowntofunctionasphagocyticreceptorforapoptoticcells.Severalinvivomodelssupporttheroleofthethrombospondin/CD36systeminangiogenesisandtumorgrowth.AnimportantroleforCD36hasbeenfoundinMalariaasmajorreceptorforP.falciparum-infectedredbloodcells.ManydifferentligandshavebeenreportedtointeractwithCD36,suggestingthatCD36couldrecognizeastructure-baseddomainratherthanspecificcontactresidues.MonoclonalantibodyCRFD-2712blocksCD36binding.Itisnotknownwhethertheantibodyisactivatingorneutralizing.TheantibodycrossreactswithratCD36.
