ThemonoclonalantibodyFA-11reactswithmurinemacrosialin(mouseCD68),aheavilyglycosylatedtransmembraneproteinof87-115kDa,whichisspecificallyexpressedbytissuemacrophages,Langerhanscellsandatlowlevelsbydendriticcells.Macrosialinbelongstothelysosomal-associatedmembraneprotein(LAMP)family.IncommonwiththeLAMPs,macrosialinisatypeIme.mbraneprotein,containingashortandhighlyconservedcytoplasmictail,followedbyatransmembranedomainthatprecedestheintraluminalregion.Inmacrophages,macrosialinismainlyexpressedasalateendosomalproteinandrapidlyexchangeswithasmallsubsetofmacrosialinpresentonthecellsurface.Severalreportshaveshownthatmacrosialinrecognisesoxidizedlow-densitylipoproteinsaswellastheintercellularadhesivemolecule(ICAM-L)raisingthepossibilityofareceptorfunctionofthisprotein.Inhuman,macrosialinhasbeensuggestedtobeanovelprognosticfactorforclassicalHodgkin’slymphoma-sincehighexpressionofmacrosialinandCD163correlateswithadverseoutcome.
ThemonoclonalantibodyFA-11detectssurfacemacrosialinatlowlevelsinresidentmouseperitonealmacrophageswhichcanbeenhancedbythiolycollatestimulation.MacrosialinispredominantlylocatedwithinthecellandcanbedetectedbyflowcytometrywiththemonoclonalantibodyFA-11whencellpermeABIlisationisused.
ThemonoclonalantibodyFA-11detectssurfacemacrosialinatlowlevelsinresidentmouseperitonealmacrophageswhichcanbeenhancedbythiolycollatestimulation.MacrosialinispredominantlylocatedwithinthecellandcanbedetectedbyflowcytometrywiththemonoclonalantibodyFA-11whencellpermeABIlisationisused.
