ThemonoclonalantibodyED31recognizesthemacrophagereceptorwithcollagenousstructure(MARCO).Thescavengerreceptors(SRs)expressedbymacrophagesarethoughttoplayanimportantroleintheimmuneresponseagainstbacteriabymediatingbindingandphagocytosis.AnimportantfamilyofreceptorsinvolvedintheseprocessesaretheclassAmacrophagescavengerreceptors(SR-A).TherearethreeclassesofSRs,A,BandC.SR-A,typeIandtypeII,arehomotrimericglycoproteinscomposedofthree77kDmonomers.MARCOisamemberofthisclassofscavengerreceptors.ThemolecularstructureofMARCOresemblesthatofSR-AI,containingatriple-helicalcollagenousdomainandascavengerreceptorcysteine-rich(SRCR)domainattheCterminus.MARCOisexpressedbydistinctpopulationsofmacrophagesinthespleenandlymphnodes.Itisrapidlyinducedonmacrophagesinothertissues(e.g.LiverKupfercells)duringinfectionorLPStreatment.MARCOseemstoplayanimportantroleinthelungsresponsetoinhaledparticlesandairbornepathogens.ThemonoclonalantibodyED31bindstotheC-terminalcysteinerichdomainofMARCO,andhasbeenshowntoblockligandbinding.
