Themonoclonalantibody2A11recognizesmouseβ-glucanreceptor,alsoknownasdectin-1.Dectin-1isasmalltypeIItransmembranereceptorcontainingonelectin-likecarbohydraterecognitiondomain.Thisdomainrecognizesβ1,3-and/orβ1,6-linkedglucans,intactyeastandanimmunoreceptortyrosine-basedactivationmotif(ITAM)inthecytoplasmictail.β-Glucanisoneofthemostabundantpolysaccharidesinfungalpathogens.Byrecognizingβ-glucan,dectin-1hasafundamentalfunctioninantifungalimmunity.Dectin-1isasignalingnon–Toll-likepattern-recognitionreceptorrequiredfortheinductionofprotectiveimmuneresponses.Dectin-1isresponsIBLefortheβ-glucan-dependent,nonopsonicrecognitionofzymosanbyprimarymacrophages.Zymosanisayeastderivedparticlecomposedprincipallyofpolysaccharides,ofwhichβ-glucanandmannanarethemajorconstituents.InvivoadmiNISTrationofzymosan,orpurifiedsolubleβ-glucans,hasanumberofdesirableeffectsonimmunefunction,includingtheABIlitytoconferresistancetotumorsandvariousinfections.ThereceptorishighlyexpressedonmacrophagesandneutrophilsandtoalesserextentondendriticcellsandasubpopulationofTcells.Dectin-1participateswiththecomplementreceptorCR3inahostproinflammatorycytokineresponsetoafungalpathogen.Themonoclonalantibody2A11recognizesanextracellularepitopeondectin-1.
