ThemonoclonalantibodyBM8recognisesa125kDextracellularmacrophagemembranemolecule,highlyrestrictedtomaturemacrophagesubpopulationsresidingintissue.ThismurineF4/80glycoproteincontainsseven-transmembrane(TM7)regions,whichanchortheproteininthecellmembrane,andtherebyshowssimilarityinthisregiontoG-protein-coupledreceptors.TheF4/80moleculesharesoverallstructuralhomologytoothermembersoftheepidermalgrowthfactor(EGF)-TM7family.Theantigenisdetectedontissuefixedmacrophagesinallorganstestedsofar(spleen,lymphnodes,thymus,liver,skin).ItisalsopresentonLangerhanscellsintheskinandKupffercellsintheliver.Itisabsentongranulocytes,lymphocytesandtrombocytes.TheexpressionofF4/80increasesuponmaturationofmacrophageprecursorsinbonemarrowandbloodaswellasinontogeny.
ThemonoclonalantibodyBM8istheonlymacrophageMarkerthatisabletodistinguishnon-destructivefromdestructiveinflammationprocessesinthepancreas.FurThermoreitisauniquehistologicalmarkeroftheprogressionfromperi-insulitistobeta-celldestructionanddiabetesinamousediabetesmodel.ThemonoclonalantibodyBM8crossreactswithhumanheartmacrophages.
