ThemonoclonalantibodySa14-2recognizesthemousemonocyteMarkerCD14.TheCD14receptorisapatternrecognitionmoleculeintheinnateimmuneresponseagainstmicroorganismsandotherexogenousandendogenousstressfactors.CD14wascharacterizedasareceptorforLPS.TheCD14geneconsistsoftwoexonswhichcodeforasinglemRNAthatistranslatedintoaproteinof375aminoacids.TheCD14proteiniscomposedofelevenleucin-richrepeats,whicharealsofoundinTLRandwhichareimportantinPAMPbinding.IncontrasttoTLR,however,CD14lacksatransmembranedomain,andthuscannotinitiateintracellularsignaltransductionbyitself.ThemostimportantCD14signalingco-receptoristoll-likereceptor4(TLR4),whichactivates,amongothers,thenuclearfactorκB(NF-κB)inflammatorypathway.TheCD14proteinisprocessedintheendoplasmaticreticulumandexpressedasa55kDaglycoproteinonthecellsurfaceviaaglycosylphosphatidyl(GPI)anchor.LikeotherGPI-anchoredproteins,CD14accumulatesonthecellsurfaceinmicrodomainsknownaslipidrafts.CD14isexpressedpredominantlyonthesurfaceof"myeloid"cells,suchasmonocytes,macrophagesandneutrophils,butatlowerlevelsalsoonepithelialcells,endothelialcellsandfibroblasts.
CD14isalsoexpressedinasolubleform(sCD14).sCD14mayresultfromsecretionoftheproteinbeforecouplingtotheGPIanchororfromsheddingorcleavagefromthesurfaceofmonocytes.sCD14ispresentinthecirculationandotherbodyfluidsandlevelsofsCD14inplasmaincreaseduringinflammationandinfection.CD14isamoleculewithabroadrangeoffunctions.Inadditiontofunctioningasapatternrecognitionreceptorforavarietyofmicrobialligands,CD14alsoactsasareceptorforendogenousmoleculeslikeintercellularadhesionmolecule(ICAM)-3onthesurfaceofapoptoticcells,amyloidpeptid,ceramide,anduratecrystals.LigationofCD14bytheseligands,exceptforapoptoticcells,mediatesactivationofinflammatoryresponses.
