MonoclonalantibodymT2.7reactswithmouseToll-likereceptor2(TLR2,CD282).Toll-likereceptors(TLR)arehighlyconservedthroughoutevolutionandhavebeenimplicatedintheinnatedefensetomanypathogens.InDrosophilatollisrequiredfortheanti-fungalresponse,whiletherelated18-wheelerisinvolvedinantibacterialdefenses.Inmammals,TLRidentifiedastypeItransmembranesignalingreceptorswithpatternrecognitioncapABIlities,havebeenimplicatedintheinnatehostdefensetopathogens.TLR2hasbeenidentifiedasareceptorthatiscentraltotheinnateimmuneresponsetolipoproteinsofGram-negativebacteria,severalwholeGram-positivebacteria,aswellasareceptorforpeptidoglycanandlipoteichoicacidandotherbacterialcellmembraneproducts.AfunctionalinteractionbetweenTLR2andTLR6inthecellularresponsetovariousbacterialproductshasbeendiscovered.ThecurrentlyacceptedparADIgmregardsTLR2asanessentialreceptorformanyeubacterialcellwallcomponents,includinglipoproteinsandpeptidoglycan.Bacterialspeciesasdiverseasmycobacteria,spiRochetes,mycoplasma,Staphylococcusaureus,andStreptococcuspneumoniaehaveallbeenshowntomediatecellularactivationviaTLR2.ThemonoclonalantibodymT2.7stainedoverexpressed,aswellasendogenouscellsurface-andintracellularTLR2.TheantibodydoesnotaffectcellactivationthroughTLR2.
