Themonoclonalantibody8F4recognizesmousemyeloperoxidase(MPO).MPOisaglycoproteinproducedasasingleprecursor,whichissubsequentlycleavedintoaalphaandbetachain.InhumantheBIOLOGicallyactiveMPOisa150kDatetramercomposedof2glycosylatedalfachainsof59-64kDaand2betachainsof14kDa.MPOisstoredinazurophilicgranulesofpolymorphonuclearleukocytesandisrapidlyreleasedintothephagosomeandextracellularspaceduringinflammatoryconditions.Theenzymecatalyzestheconversionofchlorideandhydrogenperoxidetohypochlorite,apotentoxidant,whichfunctionsinhostdefenseagainstmicroorganisms.
InvolvementofMPOhasbeendescribedinseveralhumandiseases,suchascardiovasculardisease,airwayinflammation,lungcancer,Alzheimer"sdiseaseandmultiplesclerosis.ApositivecorrelationbetweenelevatedMPOlevelsinserumandcardiovasculardiseasesuggestaninterestingroleforMPOasandiagnosticMarker,makingitpossIBLetoidentifypatientsatriskforfuturecardiacevents.FurThermore,therearesomeautoimmunediseases,inwhichMPOistargetedbyantineutrophilcytoplasmantibodies.StudieswithMPO-knockoutmicehaveshownanincreasedsusceptibilitytopneumoniafollowingintratrachealinfections.Moreover,MPOdeficientmicearemoresusceptibletoexperimentalautoimmuneencephalitis,aTcell-dependentneuronaldisease,andhaveanincreasedexpressionofarterioscleroticplaquescomparedtowild-typemice.Theanti-mouseMPOmonoclonalantibody8F4recognizesnaturalMPOinbiologicalsolutionsbyELISA,infrozentissuesectionsfixedwithacetoneandinflowcytometryusingacellpermeABIlizationmethod.
