						Description:Mannosereceptor,Mouse,mAbMR5D3
ThemonoclonalantibodyMR5D3recognizestheMannoseReceptor(MR,alsoknownasCD206),amemberofthevertebrateC-typelectinfamily.MRisapatternrecognitionreceptorthatisinvolvedinbothinnateandadaptiveimmunity.The175kDatransmembraneproteinconsistsof5domains:anamino-terminalcysteine-richregion,afibronectintypeIIrepeat,aseriesofeighttandemlectin-likecarbohydraterecognitiondomains(responsIBLefortherecognitionofmannoseandfucose),atransmembranedomainandanintracellularcarboxy-terminaltail.Thestructureissharedbythefamilyofmultilectinmannosereceptors:thephospholipaseA2-receptor,DEC205andthenovelC-typelectinreceptor(mannosereceptorX).TheMRrecognisesawiderangeofgrampositiveandgramnegativebacteria,yeasts,parasitesandmycobacteria.Moreover,theMRhasalsobeenshowntobindandinternalizetissue-typeplasminogenactivator.
MR’sarepresentonmonocytesanddendriticcells(DC)andarepresumedtoplayaroleininnateandadaptiveimmunity,thelatterviaprocessingbyDC.TheexpressionofMRasobservedinimmunohistologyispresentontissuemacrophages,dendriticcells,asubpopulationofendothelialcells,Kupffercellsandspermcells.TheexpressionofMRonmonocytesincreasesduringcultureandcanbeenhancedbycytokinessuchasIL-4.LabellingofMRexpressingmonocytes/macrophagesincreaseswithprolongedincubationtimeprobablyduetointernalizationoftheMR-antibody-complex.DetectionoftheMRwithanti-MRmonoclonalantibodyMR5D3cansubstitutestainingformannosecontainingprobesaslabeledmannosylatedBSA,atechniquewhichismorecumbersomeandlessspecific.
