Monoclonalantibody6C2reactswithmouseToll-likereceptor2(TLR2,CD282).Toll-likereceptors(TLR)arehighlyconservedthroughoutevolutionandhavebeenimplicatedintheinnatedefensetomanypathogens.InDrosophilatollisrequiredfortheanti-fungalresponse,whiletherelated18-wheelerisinvolvedinantibacterialdefenses.Inmammals,TLRidentifiedastypeItransmembranesignalingreceptorswithpatternrecognitioncapABIlities,havebeenimplicatedintheinnatehostdefensetopathogens.TLR2hasbeenidentifiedasareceptorthatiscentraltotheinnateimmuneresponsetolipoproteinsofgram-negativebacteria,severalwholegram-positivebacteria,aswellasareceptorforpeptidoglycanandlipoteichoicacidandotherbacterialcellmembraneproducts.AfunctionalinteractionbetweenTLR2andTLR6inthecellularresponsetovariousbacterialproductshasbeendiscovered.ThecurrentlyacceptedparADIgmregardsTLR2asanessentialreceptorformanyeubacterialcellwallcomponents,includinglipoproteinsandpeptidoglycan.Bacterialspeciesasdiverseasmycobacteria,spiRochetes,mycoplasma,Staphylococcusaureus,andStreptococcuspneumoniaehaveallbeenshowntomediatecellularactivationviaTLR2.
