Themonoclonalantibody16D2recognizesmousecomplementfactorC4,formerlyknownasGgprotein,whichconsistsofanalpha-,beta-,andgamma-chain.TheclassicalpathwayofcomplementandtheMannosebindinglectinactivationpathwayconvergeatC4.C1s,MASP-1andMASP-2cleaveC4resultingintheformationofC4aandC4b.Subsequently,C4bcanbecleavedtoC4candC4dbyotherserumenzymes.Themonoclonalantibody16D2reactswithintactC4,C4bandC4d.
C4isanacutephaseproteinthatisproducedbyhepatocytes,monocytesandintestinalepithelialcellsandcanbeused-inexperimentalanimalsasaMarkerforactivationoftheclassicalcomplementpathway.RecentstudieshavedemonstratedanassociationbetweengraftrejectionandC4ddepositioninamousemodelforcardiactransplantation.
C4isanacutephaseproteinthatisproducedbyhepatocytes,monocytesandintestinalepithelialcellsandcanbeused-inexperimentalanimalsasaMarkerforactivationoftheclassicalcomplementpathway.RecentstudieshavedemonstratedanassociationbetweengraftrejectionandC4ddepositioninamousemodelforcardiactransplantation.
