Themonoclonalantibody5G5reactswiththeToll-likereceptor9(TLR9,CD289).TLRsarehighlyconservedthroughoutevolutionandhavebeenimplicatedintheinnatedefencetomanypathogens.InDrosophila,tollisrequiredfortheanti-fungalresponse,whiletherelated18-wheelerisinvolvedinantibacterialdefences.Inmammals,TLRsidentifiedastypeItransmembranesignallingreceptorswithpatternrecognitioncapABIlities,havebeenimplicatedintheinnatehostdefencetopathogens.AsinvestigatedsofarallfunctionalcharacterizedTLRsignalviatheTLR/IL-1receptor(IL-1R)pathwaywhererecruitmentofMyD88seemstobeessential.Incontrasttocell-wallcomponents,bacterialDNAisprobablyinvisIBLeforimmunecellsuntilDNAisliberatedduringprocessestakingplaceintheendosomal/lysosomalcompartmentwhereintracellularTLR9recruitsMyD88toinitiatesignaltransduction.UnmethylatedCpG-dinucleotide-containingsequencesarefoundmuchmorefrequentlyinbacterialgenomesthaninvertebratesgenomes,whereasthefrequencyofCpGdinucleotidesaresuppressedandusuallymethylated.TheregionsadjacenttotheCpGdinucleotidesalsoaffecttheimmunostimulatoryactivity.Theoptimalsequencedifferssignificantlybetweenmammalianspecies.MethylatedCpGdinucleotideslackimmunostimulatoryactivities.CellularactivationinresponsetobacterialDNAandsyntheticdinucleotidescontainingunmethylatedCpG-dinucleotidesismediatedbyTLR9.Themonoclonalantibody5G5reactswithRAWmacrophagesandTLR9transfectedHEK293cells,anditiscrossreactivewithcanineTLR9.
