MannoseBindingLectin(MBL)alsocalledmannose-ormannan-bindingprotein(MBP)isamemberofthegroupofcollectins.MBLisanoligomericlectinthatrecognizescarbohydratesasmannoseandN-acetylglucosamineonpathogens.MBLcontainsacysteinerich,acollagenlikeandacarbohydraterecognitiondomain.ItformsacomplexwithC1r/C1slikeserineproteasesdesignatedMASPsthatproteolyticallycleaveC4,C2andC3.MBLisabletoactivatethecomplementpathwayindependentoftheclassicalandalternativecomplementactivationpathways.TheMBL-MASPpathway(betterknownasthelectinpathway)isantibodyandC1q-independent.MBLexhibitscomplement-dependentantibacterialactivityandactsdirectlyasanopsonicandthereforeplaysanimportantroleininnateimmunity.MBLissynthesizedbyhepatocytesandhasbeenisolatedfromtheliverorserumofseveralvertebratespecies.OnlyoneformofhumanMBLhasbeencharacterized,whiletwoformsarefoundinrhesusmonkeys,rabbits,ratsandmice.ThemouseformsareknownasMBL-AandMBL-C.TheMBL-Cconcentrationsinserumareabout6-foldcomparedtothatofMBL-A.MBL-A,butnotMBL-CwasfoundtobeanacutephaseproteinincaseinandLPS-injectionmodels.MBL-CexistsinhigheroligomericformsthanMBL-A.Themonoclonalantibody16A8isacalcium-dependentantibody.
