ThemonoclonalantibodyM330-19reactshighlyspecificwithmousenaturalandrecombinantLBP.TheantibodyisatypeIantibodyblockingtheLPSbindingtoLBP.LPSbindingprotein(LBP)isanapproximately60kDaacutephaseproteinthatisproducedbyhepatocytes.ThisproteinstronglybindstoLPSandhasbeenshowntoplayanimportantroleinthehandlingofLPSbythehost.AnumberoffunctionsofLBPhavebeenreported.First,LBPtransfersLPStotheLPSreceptorCD14onmononuclearphagocytes,leADIngtoan100-1,000-foldincreasedsensitivityofthecellstoLPS.FurThermore,LBPcanenhancetheresponseofCD14negativecellsbyaccelerationofLPSbindingtosolubleCD14,acomplexthatstimulatesthesecells.Next,LBPtransfersLPSintoHighDensityLipoprotein(HDL),whicheffectivelyneutralizesitsBIOLOGicalpotency.LBPwasdemonstratedtoprotectmicefromsepticshockcausedbyLPSorgramnegativebacteria.
