ThemonoclonalantibodyV1qrecognizesmousetumornecrosisfactoralpha(TNF-α).TNF-αistheprototypecytokineofthefamilyofTNF-relatedligands,whicharebasedonstructuralandfunctionalhomologies.TNF-αissynthesizedastypeIItransmembraneprotein.TNF-αcanberecognizedbytwodifferentmembranereceptors,namelyTNF-R1andTNF-R2.TNF-αispresentinamembrane-bound(tmTNF)aswellassolubleform(sTNF).Themembrane-boundformofTNF-αisrecognizedbybothTNFreceptorswithhighaffinity,whereasthesolubleformisrecognizedmoresuperiorlybyTNF-R1.TNF-αisproducedbymanydifferentcelltypesincludingmacrophages,Tlymphocytes,NKcells,neutrophilsandendothelialcells.CellsdifferintheexpressionofthetwoTNF-receptorsandsTNFversustmTNF,respectively.TNF-α,ahomotrimeric17kDaprotein,isapotentmediatorofinflammatoryandmetabolicfunctions.TNF-αwasoriginallydetectedasahighlycytotoxiccytokinefortumorcells,itcausestumornecrosisinvivoandshowscytolyticactivityagainsttumorcellsinvitro.FurThermore,TNF-αhasbeenimpliedascentralmediatorinshockinducedbygramnegativemicro-organisms.TNF-αinducesonitsturntheproductionofmanyothercytokines.Furthermore,TNF-αhasbeenfoundininflammatoryfocisuchassynovialeffusionsinrheumatoidarthritis,systemiccirculationinsepticshock,parasitemiaandrejectionofrenaltransplants.ThemonoclonalantibodyV1qrecognizesbothnaturalandrecombinantTNF-αandshowsneutralizingactivity.
