Themonoclonalantibody3/11recognizesC3a,anactivatedfragmentofthecomplementproteinC3,a190kDaproteinwhichisthemostabundantcomplementproteininserum.C3isthethirdcomponentofthecomplementsystemandplaysacentralroleinthecomplementcascade.Thethreepathwaysofcomplimentactivationcometogether-atthiskeymolecule.CleavageofthepivotalcomplementcomponentC3resultsinthegenerationofasetofeffectormoleculeswhichhavediverseBIOLOGicalfunctions.ThecleavageproductC3bpromotescomplementacrtivationandthesubsequentformationofthemembraneattackcomplex.C3a,thesmall(ca.10kDa)cleavageproductpossessesanaphylatoxicaswellasimmunoregulatory-properties.Itexertsitsfunctionthroughaspecificreceptor(C3aR),whichbelongstotheG-proteincoupledreceptorfamily.ExpressionofC3aR-hasbeenreportedinmanycelltypesincludingmyeloidandnon-myeloidcells.ExpressionofC3aRonhaematopoieticstem/Progenitorcellshasbeenshowntopromotethetrafficking/homingofthesecellstotheBM.EngagementofC3aRonDCsandTcellshasbeenshowntoup-regulatethesecellfunctions.
Themonoclonalantibody3/11reactsspecificallywiththeC3aanaphylatoxin,providingatoolformonitoringacutecomplementactivation(C3ageneration)inexperimentalmousemodels.
