Themonoclonalantibody266-6K1recognizeshumanmyeloperoxidase(MPO),an~135glycoproteinexpressedinallcellsofthemyeloidlinage.MPOfunctionsasanα2β2heteromultimerconsistingoftwoheavy(α)andtwolight(β)chainsof55and15kDarespectively.MPOisabundantlypresentinazurophilicgranulesofpolymorphonuclearneutrophils(PMNs).Itisanimportantenzymeusedduringphagocyticlysisofengulfedforeignparticleswhichtakespartinthedefenseoftheorganismthroughproductionofhypochlorousacid(HOCl),apotentoxidant.InthestimulatedPMN,MPOcatalyzestheproductionofhypohalousacids,primarilyhypochlorousacidinphysiologicsituations,andothertoxicintermediatesthatgreatlyenhancePMNmicrobicidalactivity.Uponactivationofneutrophils,MPOcanberapidlyreleasedandassuchusefulinbodyfluidsasMarkerforinflammatorystatus.
InvolvementofMPOhasbeendescribedinnumerousdiseasessuchasatherosclerosis,lungcancer,Alzheimer"sdisease,inflammatoryboweldiseaseandmultiplesclerosis.AutoimmuneantibodiestoMPO(socalledANCA)areinvolvedinWegener’sdisease.SincethediscoveryofMPOdeficiency,initiallyregardedasrareandrestrictedtopatientssufferingfromsevereinfections,MPOhasattractedmoreclinicalattention.
InvolvementofMPOhasbeendescribedinnumerousdiseasessuchasatherosclerosis,lungcancer,Alzheimer"sdisease,inflammatoryboweldiseaseandmultiplesclerosis.AutoimmuneantibodiestoMPO(socalledANCA)areinvolvedinWegener’sdisease.SincethediscoveryofMPOdeficiency,initiallyregardedasrareandrestrictedtopatientssufferingfromsevereinfections,MPOhasattractedmoreclinicalattention.
