Monoclonalantibody6G3reactsspecificallywithArginaseI,thefinalenzymeintheureacycle,whichisresponsIBLeforthehydrolysisofargininetoureaandornithine.Thehighestconcentrationoftheenzymeispresentintheliverinwhichthebulkofureagenesisoccurs.Twotypesofarginasesareknown:ArginaseIandII.ThecytosolicenzymefoundprimarilyinliverisArginaseI,a35kDproteinthatcirculatesastrimer.ArginaseIIisexclusivelylocatedinthemitochondrion.ArginaseIisnexttotheliverinmanalsoexpressedbymaturefetalandadultredbloodcellsandactivatedmonocyticcells.DuringinflammationinductionofArginaseIbyinflammatorycytokinesinmonocyticcellsisconsideredtoleadtoalocaldepletionofarginineresultinginamicroenvironmentthatpreventsnitricoxideproductionandargininedependentTcellfunction.ArginaseIIisexpressedbykidney,nucleatedredbloodcells,brain,spinalcord,gastro-intestinaltract,mammaryglandandprostate.EnhancedcirculatingArginaseIlevelshavebeenreportedaftersurgery,followinghaemorrhageandinasthmaticpatients.MeasurementofcirculatingArginaseIhasbeenusedexperimentallyasrapidMarkerforliverinjury.
