Themonoclonalantibody2991recognizestheneo-epitopeofhumanC3a/C3a-desArgthatisformeduponcleavageofthecomplementproteinC3intoC3aandC3b.Thecomplementsystemisanimportantpartofthehumoralresponseininnateimmunity,consistingofthreedifferentpathways.Thethirdcomplementcomponent,C3,iscentraltotheclassical,alternativeandlectinpathwaysofcomplementactivation.Activationproductsofthecomplementcascadecontainneo-epitopesthatarenotpresentintheindividualnativecomponents.Monoclonalantibodiesdetectingneo-epitopeshavebeenusedfordirectquantificationofactivationatdifferentstepsinthecomplementcascade.ThesynthesisofC3istissue-specificandismodulatedinresponsetoavarietyofstimulatoryagents.C3isthemostabundantproteinofthecomplementsystemwithserumproteinlevelsofabout1.3mg/ml.AninheriteddeficiencyofC3predisposesthepersontofrequentassaultsofbacterialinfections.Inulcerativecolitis,andidiopathicchronicinflammatoryboweldisease,thedepositionofC3inthediseasedmucosahasbeenreported.ProteolysisbycertainenzymesresultsinthecleavageofC3releasingC3aanaphylatoxinandC3b.C3aisaproteinof74aminoacids.C3aitselfisveryshort-livedandinserumiscleavedrapidlyintothemorestableC3a-desArg(alsocalledacylationstimulatingprotein,ASP).Therefore,measurementofC3a-desArgallowsreliableconclusionsaboutthelevelofcomplementactivationinthetestsamples.C3aisamediatoroflocalinflammatoryprocesses.Itinducessmoothmusclecontraction,increasesvascularpermeABIlity,andcauseshistaminereleasefrommastcellsandbasophilicleukocytes.C3aisinvolvedininflammatoryreactionsseeningram-negativebacterialsepsis,trauma,ischemicheartdisease,post-dialysissyndromeandavarietyofautoimmunediseases.Normalvaluesinplasmaofcontrolpersonsrangebetween48150ng/ml(median:86.4ng/ml,SD:29.3ng/ml)
